Description: Atilmotin, [N+-α-(CH3)3Phe1,D-Arg12,Leu13,Lys14] motilin 1–14 amide, is a synthetic peptide analog of motilin that is under development for disorders of gastrointestinal motility, particularly postoperative ileus. Preclinical pharmacological evaluation of atilmotin has shown it is a potent agonist at motilin receptors in vitro and in vivo. Unlike other motilin agonists, including erythromycin-like agents, atilmotin is short acting, with t1/2 less than 10 min. Atilmotin, at doses of 6, 30 or 60 µg intravenously, has been shown to increase gastric emptying within the first 30 min after administration. Thus, atilmotin might have clinical utility for GI motility disorders.
