Description: In the present study, we identified and characterized a novel synthetic peptide, ten amino acids long, termed Ilantide, which is derived from the human IL-Ra N-terminal domain involved in interactions with IL-1RI. We hypothesized that Ilantide would compete with IL-1R signaling, thereby inhibiting inflammatory responses induced by various conditions in vitro and in vivo. Indeed, we found that Ilantide bound to IL-1RI and inhibited the IL-1-induced activation of NF-κB and secretion of tumor necrosis factor α (TNF-α) by macrophages. It protected pancreatic islets from IL-1β-induced apoptosis and reduced inflammation in collagen-induced arthritis (CIA). The peptide penetrated the BBB, ameliorated decline in social activity and memory in lipopolysaccharide (LPS)- and amyloid-treated animals and delayed the development of experimental autoimmune encephalomyelitis (EAE).
