Description: S-Ac-MAG3-NHS, a modified mercaptoacetyltriglycine (MAG3) chelator, which has acetyl S-protection and C-terminal derivitized with N-hydroxysuccinimide (NHS) ester for conjugation, has been used to radiolabel four small peptides, bovine pancreatic trypsin inhibitor, epidermal growth factor, human neutrophil elastase inhibitor and plasmin inhibitor, with 99mTc. The mercaptoacetyltriglycine (MAG3) chelator has been shown to stably complex technetium-99m (99mTc) for nuclear imaging and radiorhenium (186/188Re) for tumor radiation therapy studies. The bifunctional N-hydroxysuccinimidyl ester of MAG3 with S-acetyl protection (N-hydroxysuccinimidyl S-acetylmercaptoacetyltriglycinate (NHS-MAG3)) has been successfully used to covalently conjugate a MAG3 chelator to primary amine functionalized biomolecules. The use of NHS-S-acetyl-MAG3 may be a convenient method of radiolabeling peptides with 99mTc.
