Description: The tripeptide thyrotropin releasing hormone (TRH; L-pyroglutamyl-L-histidyl-L-prolineamide (L-pGlu-L-His-L-ProNH2)) is primarily known for its role as the central regulator of the hypothalamic-pituitary-thyroid axis (HPT axis). The HPT axis plays a crucial role in regulating and maintaining somatic metabolism, thermogenesis, blood pressure, core body temperature, respiration rate, and food and water intake. TRH is distributed throughout the brain and the periphery; reports describe the presence of TRH in the hypothalamus, pituitary, brainstem, cerebellum, cortex, thalamus, hippocampus, as well as the pancreas, gastrointestinal tract, and spinal cord. TRH, like melatonin, is an anti-aging agent with a broad spectrum of activities that, because of their actions, suggest that TRH has a fundamental role in the regulation of metabolic and hormonal functions. TRH and its synthetic analogs have been recognized as trophic factors in neurons of the diencephalon and spinal cord, and as neuroprotectants against oxidative stress, glutamate toxicity, caspase-induced cell death, DNA fragmentation, and inflammation.
