Description: Leu-Leu and Leu-Leu-Leu have been utilized to formulate spray‐dried pulmonary formulations with improved aerosol performance (dispersibility) and stability. Trileucine-containing particles are corrugated, owing to trileucine’s low aqueous solubility. This result in significantly improved aerosol performance. However, corrugation is not the only factor for improving aerosol performance, which seems to be also influenced by the low surface energy, noncohesive nature of trileucine particles, which is a reflection of its surface activity in solution. Finally, peptide and protein aggregation and chemical degradation (deamidation and hydrolysis) can be reduced by the addition of trileucine in the formulation, presumably related to trileucine surface activity in solution.
